Resumen

Sus antecedentes:

  • El di(2-etilhexil) ftalato (DEHP) es un contaminante ambiental generalizado relacionado con problemas reproductivos en mujeres.
  • Estudios previos indican que la exposición a DEHP agota los folículos ováricos y altera la producción hormonal.
  • Los mecanismos moleculares precisos que subyacen al impacto del DEHP en la función ovárica siguen siendo en gran medida desconocidos.

Objetivo del estudio:

  • Investigar los efectos in vivo del DEHP en el desarrollo y la función ovárica en ratones hembra.
  • Elucidar las vías moleculares implicadas en el daño ovárico inducido por DEHP.

Principales métodos:

  • Se expuso un modelo de ratón in vivo a 200 mg/kg de DEHP durante 28 días.
  • Se evaluó el desarrollo ovárico, el recuento de folículos, la dinámica mitocondrial, el estrés oxidativo y el daño al ADN.
  • Se analizaron vías de señalización clave, incluidas Sirt3/Sod2 y Akt/mTOR.

Principales resultados:

  • La exposición a DEHP inhibió el desarrollo ovárico y la maduración folicular, reduciendo el número de folículos primarios y antrales.
  • Se alteró la dinámica mitocondrial, lo que provocó mitofagia y autofagia.
  • El DEHP indujo estrés oxidativo, metabolismo energético mitocondrial anormal, daño al ADN y apoptosis.
  • Se observó la inhibición de las vías de señalización Sirt3/Sod2 y Akt/mTOR.

Conclusiones:

  • La exposición a DEHP perjudica significativamente la función ovárica y la capacidad reproductiva femenina.
  • Los efectos perjudiciales se deben a la inhibición de las vías de señalización Sirt3/Sod2 y Akt/mTOR.
  • Esta investigación proporciona información crítica sobre los mecanismos moleculares de la toxicidad del DEHP en el sistema reproductivo femenino.
