Sus antecedentes:

  • El microambiente tumoral (TME) influye críticamente en la progresión del cáncer, la evasión inmune y la resistencia al tratamiento.
  • Comprender la dinámica del TME es vital para mejorar los resultados de los pacientes y desarrollar terapias efectivas.
  • Los modelos in vitro existentes a menudo no logran capturar la complejidad del TME.

Objetivo del estudio:

  • Ingenierizar un sofisticado modelo tumoral 3D in vitro que recapitule el TME in vivo.
  • Proporcionar una plataforma biomimética para estudiar ecosistemas tumorales complejos y facilitar el descubrimiento de fármacos.

Principales métodos:

  • Ingenierizaron constructos tumorales 3D de alta densidad a escala milimétrica.
  • Incorporaron la comunicación bidireccional entre células cancerosas, fibroblastos asociados al cáncer (CAF) y macrófagos asociados a tumores (TAM).
  • Integraron una red vascular jerárquica con estructuras venosas y similares a capilares.

Principales resultados:

  • Los constructos 3D imitaron con éxito los sellos distintivos arquitectónicos y funcionales del TME in vivo.
  • Demostraron la diferenciación inducida de CAF y TAM a través de interacciones con células cancerosas.
  • Modelaron con éxito distintos fenotipos inmunes de tumores 'calientes' y 'fríos'.
  • Replicaron una red vascular jerárquica dentro del modelo tumoral.

Conclusiones:

  • El constructo tumoral 3D desarrollado sirve como un modelo in vitro robusto y preciso del TME in vivo.
  • Esta plataforma tiene un potencial significativo para avanzar en la investigación del cáncer, particularmente en la comprensión de los ecosistemas tumorales y la aceleración del descubrimiento de fármacos.
