    Sus antecedentes:

    • Thy-1 es una glicoproteína de la superficie celular involucrada en las interacciones célula-célula.
    • La glicosilación es una modificación post-traduccional crítica que afecta la función de las proteínas.
    • Grupo de complementación Clase E Los linfomas Thy-1-mutantes presentan un defecto específico de glucosilación.

    Objetivo del estudio:

    • Para identificar el defecto específico de glucosilación en los linfomas tipo E Thy-1-mutantes.
    • Para caracterizar los oligosacáridos ligados a lípidos (LLOs) en estas células mutantes.
    • Para determinar el origen de oligosacáridos anormales encontrados en polipéptidos recién sintetizados.

    Principales métodos:

    • Aislamiento y caracterización de LLOs de células de linfoma de tipo salvaje y mutantes de Thy-1.
    • Análisis de LLOs utilizando la digestión de la endoglucosidasa H.
    • Análisis de oligosacáridos en polipéptidos recién sintetizados a partir de células mutantes.

    Principales resultados:

    • Se aislaron dos LLOs principales de células mutantes, ambos más pequeños que los LLOs de tipo salvaje.
    • Los LLOs mutantes eran resistentes a la digestión de la endoglucosidasa H, a diferencia de los LLOs de tipo salvaje.
    • Los oligosacáridos en los polipéptidos recién sintetizados en células mutantes también fueron resistentes a la endoglucosidasa H.

    Conclusiones:

    • El defecto de glicosilación en los linfomas de clase E Thy-1 es un bloqueo en la síntesis de precursores de LLO.
    • Los LLOs anormales, resistentes a la endoglucosidasa H, se forman en células mutantes.
    • Estos LLOs anormales son la fuente probable de los oligosacáridos resistentes a la endoglucosidasa H en las glicoproteínas de las células mutantes.
