    Sus antecedentes:

    • Myxococcus xanthus es una bacteria gram-negativa.
    • La bacteria contiene 500-700 copias por cromosoma de un fragmento de ADN lineal corto de una sola hebra.

    Objetivo del estudio:

    • Para caracterizar el nuevo fragmento de ADN encontrado en Myxococcus xanthus.
    • Para determinar la secuencia y estructura de este elemento único de ADN.

    Principales métodos:

    • Etiquetado del fragmento de ADN (ADN multicopia de una sola hebra; msADN) en el extremo 5'.
    • La hibridación de msDNA etiquetado a las manchas de ADN cromosómico total.
    • Clonado y secuenciación de ADN cromosómico hibridado.
    • Secuenciación directa de msDNA utilizando el fragmento de ADN polimerasa I Klenow y la transferasa deoxinucleotidilo terminal.

    Principales resultados:

    • msADN se hibridó a bandas únicas de alto peso molecular en manchas cromosómicas.
    • La porción de ADN de msDNA se compone de 163 bases.
    • La secuencia de msDNA mostró una correspondencia exacta con una secuencia cromosómica clonada.
    • Se descubrió que el extremo 5' del msADN está primado por un segmento corto de ARN.
    • Se postula una estructura secundaria elaborada para la secuencia de ADN ms.
    • Se identificó un ADN satélite similar en Stigmatella aurantiaca.

    Conclusiones:

    • Myxococcus xanthus posee un elemento único de ADN multicopia de una sola hebra (ADNms).
    • msADN se deriva del ADN cromosómico y exhibe una estructura secundaria compleja.
    • La presencia de ADN similar en Stigmatella aurantiaca sugiere que esta puede ser una característica conservada en las mixobacterias.
