Reporter genes are a type of protein-coding gene that are often tagged to a gene of interest. Once inside a target cell, reporter genes usually produce visually identifiable characteristics like fluorescence and luminescence when expressed along with the gene of interest. Thus, reporter genes “report” the presence or absence of genes of interest in an organism, determine the gene expression pattern, or track the physical location of a DNA segment or protein in the cell.

    Sus antecedentes:

    • El factor de crecimiento transformador alfa (TGF-alfa) está implicado en el desarrollo de tumores humanos.
    • El TGF-alfa puede inducir una transformación reversible en líneas celulares no transformadas.

    Objetivo del estudio:

    • Para aislar y caracterizar el gen responsable de la producción de TGF-alfa.
    • Para investigar las propiedades funcionales del TGF-alfa. maduro.

    Principales métodos:

    • Utilizó desoxiloligonucleótidos sintéticos como sondas para aislar un exón TGF-alfa de una biblioteca de ADN genómico humano.
    • Se examinó una línea celular de carcinoma de células renales humanas para el ARNm TGF-alfa utilizando el exón aislado.
    • Clonó el cDNA TGF-alfa de la línea celular identificada y lo expresó en E. coli.

    Principales resultados:

    • Se aisló con éxito un exón humano de TGF-alfa e identificó una línea de carcinoma de células renales que produce mRNA de TGF-alfa.
    • Un ADNc clonado de TGF-alfa codificaba un precursor de 160 aminoácidos.
    • El TGF-alfa de 50 aminoácidos maduro se une al receptor del factor de crecimiento epidérmico e induce la independencia de anclaje en las células normales.

    Conclusiones:

    • El estudio aisló y caracterizó con éxito el gen humano TGF-alfa y su producto.
    • El TGF-alfa maduro juega un papel importante en la transformación celular y la independencia de anclaje a través de la vía del receptor del factor de crecimiento epidérmico.
