In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA.

The endoplasmic reticulum (ER) of pancreatic β-cells synthesizes preproinsulin, which consists of a signal peptide, A and B chains, and a C-peptide. Preproinsulin is then cleaved and folded into proinsulin, which translocates to the Golgi apparatus for sorting and packaging into secretory granules.     Sus antecedentes:

    Objetivo del estudio:

    • Para construir plásmidos bacterianos recombinantes que contengan ADN complementario al ARN mensajero de la preproinsulina humana.
    • Determinar la secuencia de codificación completa y las regiones flanqueantes de la preproinsulina humana.
    • Para predecir la secuencia de aminoácidos del péptido de señal de la preproinsulina humana.

    Principales métodos:

    • Construcción de plásmidos bacterianos recombinantes.
    • Utilizando ADN complementario (ADNc) derivado del ARN mensajero de la preproinsulina humana (ARNm).
    • Empleando la secuenciación enzimática de terminación de la cadena con ARNm de insulinoma y clonaciones de ADN.

    Principales resultados:

    • Se construyó con éxito un clon de plásmido bacteriano que contiene toda la región codificante de la preproinsulina y la región 3' no traducida.
    • Se identificaron ocho nucleótidos de la región 5' no traducida en el clon.
    • La secuenciación de terminaciones de la cadena enzimática proporcionó información adicional sobre la región 5' no traducida.
    • Los resultados confirmaron la secuencia de aminoácidos previamente determinada de la proinsulina humana.
    • Se predijo la secuencia de aminoácidos del péptido de señal de la preproinsulina humana.

    Conclusiones:

    • El estudio generó con éxito una secuencia completa de ADN para la preproinsulina humana.
    • Los hallazgos validan los datos existentes sobre la secuencia de proinsulina y proporcionan nuevos conocimientos sobre el péptido de señal.
