    Sus antecedentes:

    • Las mitocondrias de la levadura contienen varias especies de ARN.
    • Las formas estructurales de estos ARN no se comprenden completamente.
    • Se han observado moléculas circulares de ARN en otros sistemas.

    Objetivo del estudio:

    • Para investigar la presencia y las características de las moléculas circulares de ARN en las mitocondrias de la levadura.
    • Para determinar las propiedades estructurales y las funciones potenciales de estos ARN circulares.

    Principales métodos:

    • Aislamiento de las fracciones de ARN mitocondrial de levadura (11S y 18S) utilizando electroforesis en gel de agarosa.
    • Microscopía electrónica para la visualización y medición de moléculas circulares de ARN.
    • Experimentos de hibridación de ARN-ADN utilizando fragmentos específicos de ADN mitocondrial.
    • Tratamientos enzimáticos (DNAasa, pronasa) y tratamientos químicos (glioxal) para evaluar la integridad estructural.

    Principales resultados:

    • Aproximadamente el 50% de las fracciones de ARN mitocondrial de levadura 11S y 18S aisladas existen como moléculas circulares.
    • Los círculos 11S tienen una longitud de contorno de 0,36 ± 0,02 micras, mientras que los círculos 18S tienen un promedio de 0,78 ± 0,11 micras.
    • El ARN circular 11S forma bucles R estables e híbridos ARN-ADN con la región oxi 3 del ADN mitocondrial, lo que sugiere un papel en la codificación de la subunidad 1 de la citocromo oxidasa.
    • Tanto los ARN circulares 11S como 18S son resistentes a los tratamientos enzimáticos y químicos, lo que indica que son estructuras covalentemente cerradas.

    Conclusiones:

    • El ARN mitocondrial de la levadura contiene una proporción significativa de moléculas circulares covalentemente cerradas.
    • Estos ARN circulares, particularmente la fracción 11S, están asociados con la región del gen oxi 3.
