    Sus antecedentes:

    Objetivo del estudio:

    • Para determinar la secuencia completa de nucleótidos del ARN mensajero alfa-globina de conejo (ARNm).
    • Para validar la precisión del ARNm secuenciado a través de la comparación con los datos existentes y la secuenciación de ARN complementario (ARNc).
    • Para analizar las características generales de la secuencia de ARNm, incluyendo la composición de las bases y las preferencias de los codones.

    Principales métodos:

    • Secuenciación de ADN utilizando el método Maxam y Gilbert.
    • Análisis del plásmido quimérico pHb 72 que contiene la inserción de ADN alfa-globina de conejo.
    • Comparación de la secuencia de ARNm obtenida con datos publicados anteriormente para los extremos 5' y 3'.
    • Utilizando un nuevo programa informático para analizar los arreglos de emparejamiento de bases.

    Principales resultados:

    • Se determinó la secuencia primaria completa del ARNm alfa-globina de conejo, que abarca la región 5' no traducida a través de los primeros 361 nucleótidos traducidos.
    • La secuencia determinada mostró un acuerdo completo con las secuencias superpuestas previamente reportadas.
    • Los resultados reafirmaron la alta fidelidad de las copias de genes sintetizadas in vitro, la clonación bacteriana y el mantenimiento.

    Conclusiones:

    • Se ha establecido con alta fidelidad la secuencia completa de ARNm alfa-globina de conejo.
    • Los hallazgos apoyan la fiabilidad de la síntesis, la clonación y el mantenimiento de copias de genes in vitro.
    • El análisis de la secuencia proporciona datos valiosos para comprender la expresión génica, el uso del codón y la estructura del ARNm.
