Resumen
El gen SUC2 en la levadura produce dos formas de invertasa, reguladas por los niveles de glucosa a nivel de ARN. Diferentes ARN del gen SUC2 se correlacionan con la producción de invertasa secretada versus la producción intracelular.

Sus antecedentes:
- El gen SUC2 en Saccharomyces cerevisiae codifica dos enzimas invertasa distintas: una forma secretada y glicosilada regulada por la represión de glucosa, y una forma intracelular no glicosilada producida constitutivamente.
- Estas dos formas de enzimas son cruciales para el metabolismo del carbono de la levadura, particularmente en la utilización de la sacarosa.
Objetivo del estudio:
- Investigar los mecanismos moleculares subyacentes a la síntesis diferencial y la regulación de las dos formas de invertasa codificadas por el gen SUC2.
- Para determinar si la regulación de la glucosa de la síntesis de la invertasa se produce a nivel del ARN.
Principales métodos:
- La clonación del gen SUC2.
- Análisis de manchas del norte para detectar y cuantificar diferentes especies de ARN.
- Análisis de la expresión génica en cepas de levadura de tipo silvestre y mutantes (por ejemplo, SNF1 locus mutantes) bajo diferentes condiciones de glucosa.
Principales resultados:
- El gen SUC2 produce dos ARN distintos (1.8 y 1.9 kb) que difieren en sus extremos 5'.
- El ARN de 1.9 kb se correlaciona con la producción de invertasa secretada y está regulado por la glucosa, mientras que el ARN de 1.8 kb se correlaciona con la invertasa intracelular y se produce constitutivamente.
- Las mutaciones en el locus SNF1 afectan la desrepresión de la invertasa secretada y la producción del ARN de 1.9 kb.
- Un alelo silencioso SUC2 produce ambos ARN bajo la regulación normal de la glucosa.
Conclusiones:
- La regulación de la glucosa de la síntesis de la invertasa en la levadura está, al menos en parte, mediada a nivel del ARN.
- Los dos ARN SUC2 se originan en diferentes sitios de inicio de la transcripción, lo que dicta la síntesis de la invertasa secretada o intracelular.
- Un modelo que propone la iniciación diferencial de ARNm explica la síntesis y regulación de ambas formas de invertasa.
