- マカクはSHIVAD8-EOで挑戦され,2週間の2つの強力なbNAbs (3BNC117および10-1074) を受けました.
- ウイルス量,CD4+ T細胞数,T細胞応答をモニターした.
- 抗レトロウイルス併用療法 (cART) を受けたマカカと比較した.
主要な成果:
- bNAbの早期投与は56~177日間検出できないウイルス性血症を引き起こした.
- 治療を受けた13匹のマカクのうち6匹は 検知不能のプラズマウイルス負荷を持つ コントローラーになった.
- さらに4匹のマカクは2年以上CD4+T細胞数と低ウイルス血症を維持し,T細胞の枯渇によりウイルスが再発した.
結論:
- 急性SHIV感染時の被動免疫療法は,強力なCD8+T細胞免疫の出現を促進する.
- このT細胞免疫は,中断時にリバウンドウイルス病を引き起こすcARTとは異なり,ウイルスの複製を持続的に制御します.
- 早期のbNAb介入は,レンチウイルス感染症の長期的な制御を達成するための潜在的な戦略です.
