  • 血液不全のヒト心臓における候補遺伝子の検証と,ATAC配列解析によるクロマチンのアクセシビリティの評価.

主要な成果:

  • 酸化性リン酸化と脂肪酸代謝のダウンレギュレーションを含む強いストレス反応プログラムを持つトランスクリプション的に明確な境界地帯を特定しました.
  • NPPB,ANKRD1,DES,UCHL1,JUN,FOXP1を含む人間の心臓に保存された境界地帯マーカーを発見した.
  • Nppb欠乏したマウスは心臓発作後に致命的な心不全を示し,Nppbの重要な役割を強調した.

結論:

  • ボーダーゾーン心筋細胞は,ホメオスタティックMEF2駆動プログラムからAP-1駆動傷害誘発プログラムにシフトします.
  • この保存された転写スイッチは,心臓発作後の急性心不全の予防に不可欠なNppb発現を含んでいます.
