研究 の 目的:

  • 空間的に解明された 単細胞のトランスクリプトミクスの脳アトラスを 作るため
  • 年齢に関連したトランスクリプトミックの変化と細胞間の相互作用を特定するための空間的老化時計を開発する.
  • 老化と若返りの介入 (運動,部分的な再プログラム) が脳細胞に与える影響を調査する.

主な方法:

  • 20歳から400万個の脳細胞を集めた スペース・トランスクリプトミクス・アトラスを作成した.
  • 機械学習ベースの空間的老化時計を開発し,トランスクリプトミックの指紋を分析しました.
  • 細胞間近性の効果を評価するために ディープラーニングを活用した.

主要な成果:

  • 珍しい細胞タイプを含む老化,若返り,病気の特定された空間的および細胞タイプ特有のサイン.
  • 神経幹細胞は若返り効果を発揮する一方で 浸透したT細胞は老化防止効果を発揮することを発見しました
  • これらの近接効果の潜在的な媒介者を特定した.

結論:

  • T細胞や 神経幹細胞のような 希少な細胞タイプは 老化中の隣接する細胞に 大きく影響します
  • これらの細胞タイプをターゲットにすることで 組織の老化を防ぐ戦略が生まれます
  • 空間的な老化時計は 細胞間の相互作用を研究し 介入を評価するための 拡張可能なツールを提供します
