主な方法:
- 雌のC57BL6/JUnibマウスは,静止状態 (SD),訓練 (TR),肥満静止状態 (OB-SD),肥満訓練 (OB-TR) の4つのグループに分けられました.
- 血清のバイオマーカー (グルコース,レプチン,TNF-α,マロンディアルデヒド),tPVAT形態 (脂質滴の大きさ,アディポネクチン含有量),および血管β- ギャラクトシダース活性について評価した.
- セロトニン (5-HT) に対するtPVATの抗収縮反応を評価した.
主要な成果:
- 肥満の静止状態のマウスは血清グルコース,レプチン,TNF-α,マロンディアルデヒドが増加し,tPVATの脂質滴が増加し,アディポネクチンが減少した.
- 肥満の静止状態のマウスは,血管β-ガラクトシダースの活性が増加し,tPVATの5HTに対する抗収縮反応が変化した.
- 肥満マウスにおける有酸素運動訓練 (OB- TR) は血清レプチン,TNF-α,マロンディアルデヒドを減少させ,tPVATの脂質滴を減少させ,アディポネクチンを増加させ,β- ガラクトシダース活性を弱め,5- HTに対する血管応答を改善した.
結論:
- 肥満は,血管老化,炎症,酸化ストレス,抗収縮機能の異常によって特徴づけられる tPVAT の有意な表型変化を引き起こす.
- エアロビックトレーニングは,肥満によるtPVATのいくつかの変化を効果的に軽減します.
- 運動はtPVATの抗収縮反応を部分的に回復させ,肥満の雌マウスの主要な循環性炎症および酸化ストレスマーカーを減少させます.
