主要な成果:
- CD5阻害は,増殖に影響を与えることなく,Th17細胞によるIL- 17A,IFN- γ,GM- CSFの産生を減少させた.
- CK2阻害はIL- 17AとGM- CSFの産生を低下させ,T細胞の増殖を阻害した.
- 両方の治療はSTAT3のリン酸化を減少させ,CD5の阻害はTh17媒介の細胞毒性を排除した.
- MS血清とCSFにおける炎症性免疫プロファイルと相関する.
結論:
- CD5-CK2-STAT3シグナリング軸は,ヒトのTh17細胞の炎症反応に不可欠です.
- CD5発現は,MSにおける炎症と細胞損傷と関連しています.
- CD5シグナル伝達経路を標的にすることは,MSやその他の自己免疫疾患の治療戦略として有望です.
