研究 の 目的:

  • 鉄のパラメータと鉄調節遺伝子発現を,ベータ・タラセミアとPVの患者で分析する.
  • これらのパラメータを健康な対照群と比較する.

主な方法:

  • 鉄のパラメータの生化学分析
  • 鉄の調節遺伝子の遺伝子発現分析
  • 研究には,輸血依存性タラセミア (TDT),非輸血依存性タラセミア (NTDT),PVの患者および健康な対照群が含まれていた.

主要な成果:

  • 両疾患における溶性トランスファーリン受容体 (sTfR) の上昇は,エリトロポエティック活動の増加を示唆する.
  • 血清フェリチンの濃度はNTDTと比較してベータ・タラセミア・メジャーで有意に高かったが,PVでは低かった.
  • ヘプシディンの濃度はTDTではフェリチンに比べて低く,PVではエリトロポエーシスの増加により低下した.
  • タラセミア網膜細胞におけるTFRC発現の減少は,鉄の吸収の低下を示し,PVはTFRCとFPN1B発現の増加を示した.

結論:

  • 骨髄への鉄の輸送を決定的に影響する.
  • 血液中の鉄分調節が調節され,全身の鉄分濃度に影響する.
  • ベータ・タラセミアとPVの間には 明確な鉄の調節不全パターンが存在する.
