- 新型抗発作薬 (ASM) のセノバマート (CNB) は,焦点性の治療に有効であることが示され,ナトリウム電流とGABAA受容体を調節することによって作用する.
研究 の 目的:
- レノックス・ガストー症候群 (LGS) の患者における補助的なセノバマート (CNB) の有効性と耐性を評価する.
主な方法:
- 16人のLGS患者 (8人の小児,8人の成人) を対象とした単一センター試験.
- 発作の頻度減少,発作の持続時間,治療による有害事象 (TEAE) について収集されたデータ
- 治療後の電気脳波 (EEG) 変化は,患者のサブセットで評価された.

- 75%の患者は発作頻度 (RR) を50%以上,31%は100%の減少を達成しました.
- 発作が6ヶ月以上続く患者は19%で,1年以上続く患者は13%でした.
- 一般的なTEAEには,眠気やアタキシアが含まれており,63%がポジティブな副作用を報告した. 3人の患者はより高い投与量で発作が悪化し,投与量を下げると回復した.
結論:
- 添加剤であるセノバマート (CNB) は,LGS患者において有望な効果と良好な耐受性を示しています.
- CNBは発作制御を改善し,LGSの認知,行動,気分に好影響を与える可能性があります.
- LGSにおけるセノバマートの予期的なランダム化比較試験によるさらなる調査が必要である.
