    • RNAシーケンシング(RNA-seq)により遺伝子活性化パターンが同定された。
    • 機械学習モデル(MLM)によりPCD融合タンパク質の結合が予測された。

    主要な成果:

    • PCD融合タンパク質の結合は、エンハンサーおよびクロマチン遷移領域におけるH3K27me3、H4K20me1、またはH3K36me3の濃縮によって予測された。
    • PCD融合タンパク質が濃縮されたエンハンサーがSRAによって誘導された遺伝子は、特異的なプロモーター特徴を示した。
    • これらの特徴には、バイバレントなヒストン修飾および転写抑制因子RESTとMTA1の濃縮が含まれていた。

    結論:

    • SRA応答性は、H3K27me3を超えた特定のクロマチンシグネチャーに依存する。
    • 分化細胞におけるエピジェネティックに抑制された領域は、レギュラトリープラスティシティを保持している。
    • SRAは、誘導性クロマチン特徴を解析するための強力なツールである。
