  • 健康な高齢者54名を対象に、14日間のアクチグラフィーと採血を実施した。血漿サイトカインはAlamar NULISASeq Inflammationパネルを用いて分析した。アクチグラフィー指標(睡眠効率、持続時間、リズムの規則性/変動性)と60の選択されたサイトカインとの関連を線形回帰で検討した。

主要な成果:

  • 睡眠と概日リズムの障害は、炎症性サイトカインプロファイルの亢進と関連していた。睡眠時間の短縮はIL-1βの上昇と関連し、睡眠効率の低下はFGF23の上昇と関連していた。リズムの規則性はIL-5RAと、リズムの断片化はCSF2RBと関連し、振幅の高さはIL-6の低下と関連していた。IL-1β、IL-6、IL-5RA、FGF23、CSF2RBの関連は、p-tau217で調整した後も有意であり、アルツハイマー病の病態とは独立していた。

結論:

  • 健康な高齢者において、睡眠障害と24時間リズムは、炎症促進性サイトカインの上昇と抗炎症性サイトカインの低下に関連していた。これらの所見は、睡眠障害と加齢における炎症を関連付ける仮説を支持する。さらなる研究では、睡眠と炎症の相互作用が高齢者の認知機能に与える影響を検討すべきである。
