研究 の 目的:

  • マウスモデルにおける記憶および行動に対する脳由来細胞外小胞(EV)の影響を調査すること。アルツハイマー病(AD)モデルからのEVが認知障害を引き起こすかどうかを判断すること。

主な方法:

  • 対照群およびAD患者の死後脳組織、ならびにAPP/PS1マウスから細胞外小胞(EV)を単離した。これらのEVを野生型(WT)マウスの海馬に定位注入した。注入後の様々な時点で行動試験を用いて認知機能を評価した。

主要な成果:

  • 対照群脳由来のEVを注入されたマウスは、運動または行動障害を示さなかった。逆に、AD脳由来のEVを受けたマウスは、有意な記憶障害を示した。これらの認知障害は、注入後14日および21日で明らかになり、AD-EVおよびAPP/PS1-EVが記憶喪失を引き起こすことを確認した。

結論:

  • 細胞外小胞(EV)は、AD関連の病理学的分子を伝達し、疾患様記憶障害を引き起こすことができる。これらの発見は、EVがADの病態生理および進行における重要な要因であることを強調する。EVは、アルツハイマー病のような神経変性疾患の有望な治療標的を表す。
