研究 の 目的:

  • APPのC末端を標的とする新規抗体を生成・特性評価すること。ヒトAD脳におけるAPP-CTF蓄積の神経解剖学的分布と形態学的特徴を調査すること。APP-CTF蓄積がADの神経病理学的特徴を表すかどうかを判断すること。

主な方法:

  • APPのC末端に対する新規マウス抗体(APP1B)を生成した。ダウン症(DS)胎児由来の初代ヒト皮質細胞培養およびAPP-C99を発現する細胞株を用いて抗体の特異性を検証した。AD(孤発性、常染色体優性、DS-AD)の個人、対照群、その他の神経変性疾患の死後ヒト脳サンプルに対して免疫組織化学を行った。

主要な成果:

  • 新規APP1B抗体は、細胞培養においてAPP-CTFを特異的に認識した。ADの影響を受けやすい脳領域において、異なるタイプのAD患者で、細胞内の「タングル様」神経APP(iAPP)の免疫染色が広範に観察された。iAPP病理は対照群、ADを伴わないDS、その他の神経変性疾患では認められず、タウやユビキチンとは共局在しなかった。

結論:

  • 細胞内APP C末端フラグメント(iAPP)病理は、アルツハイマー病の新たな神経病理学的特徴として提唱される。iAPP病理の地形学的分布と選択的な神経関与は、APPのホメオスタシス異常とタウ凝集との関連を示唆する。この発見は、AD発症の根底にある分子メカニズムに新たな洞察を提供する。
