17βエストラジオール(E2)は、アミロイドβ(Aβ)誘発性の神経炎症とミトコンドリア機能障害を軽減することにより、アルツハイマー病(AD)病理から保護する。E2治療は代謝調節因子を回復させ、神経機能を強化し、ADの治療可能性を示唆している。

  • ウェスタンブロッティング、MTT、LDH、ELISA、およびSeahorse XFアッセイを用いて、タンパク質発現、細胞生存率、細胞毒性、炎症マーカー、およびミトコンドリア機能を評価した。
  • NF-κB活性化およびpAMPKおよびPGC-1αなどの代謝調節因子のレベルを分析した。

結論:

  • E2は一次大脳皮質ニューロンにおけるAβに対して有意な神経保護効果を示す。
  • E2はAβ誘発性のミトコンドリア機能障害、炎症、および神経毒性を軽減する。
  • E2はミトコンドリア機能障害および神経炎症を標的とすることにより、アルツハイマー病の治療薬としての可能性を秘めている。
