研究 の 目的:

  • 親由来DNAメチル化インプリントが哺乳類のインプリント領域における非同期複製を制御するかどうかを調査すること。
  • これらの領域内の複製タイミングとクロマチン組織に対する長鎖非コードRNA(lncRNA)およびDNAメチル化の影響を決定すること。

主な方法:

  • モノクローナルおよびハイブリッドマウス胚性幹細胞(ESC)でゲノムワイドおよび座位特異的アッセイが実施された。
  • 解析には、複製タイミング、DNAメチル化パターン、3Dクロマチン構造、および遺伝子発現が含まれた。
  • 特定の遺伝子およびlncRNAの機能的役割を評価するために、変異マウス系統が生成された。

主要な成果:

  • マウスES細胞におけるDlk1-Dio3およびSnrpnインプリント領域で、顕著な親由来依存的な複製タイミングの非同期性が観察され、神経分化により消失した。
  • Dlk1-Dio3領域における非同期複製を媒介する差次的DNAメチル化が同定された。
  • lncRNA Meg3がDlk1-Dio3領域内の母系染色体上の早期複製を制御することが判明した。複製タイミングとTAD組織との間に関連は見られなかった。

結論:

  • Meg3によって例示される親由来DNAメチル化インプリントとlncRNA発現は、インプリント領域における複製タイミング非同期性の重要な調節因子である。
