  • RA-ILDにおける肺Tph細胞と関節液Tph細胞を比較すること。

主な方法:

  • RA-ILD患者、対照群、CTD-ILD患者の肺摘出検体の単一細胞RNAシーケンシング。
  • T細胞サブクラスターと公開されている関節液T細胞データの統合解析。
  • 細胞の検証のための肺切片の免疫蛍光染色。

主要な成果:

  • 184,814個のプロファイルされた細胞から18個の異なる細胞クラスターを同定しました。
  • RA-ILD肺では肺胞I型細胞が減少し、基底細胞/線毛細胞が増加していることが観察されました。
  • RA-ILD肺でのみ末梢ヘルパーT細胞(Tph)の存在を発見し、それらは関節液Tph細胞と比較して、活性化のシグネチャーが異なり、細胞傷害性/疲弊のシグネチャーが低いことが示されました。

結論:

  • 詳細なトランスクリプトーム解析により、RA-ILD肺における主要な細胞の違いが明らかになりました。
  • 末梢ヘルパーT細胞(Tph)はRA-ILD肺の特徴です。
  • 組織間の比較により、臓器特異的なTph細胞の特徴が強調されます。
