- 差次的遺伝子発現分析のために探索的RNAシーケンシング(RNAseq)を実施しました。

- GABA合成と放出に関与する2つの遺伝子(GAD1およびSLC32A1)は、分析されたすべての核でOCD被験者でダウンレギュレーションされました。
- カリウムチャネル遺伝子KCNN3はOCD被験者でアップレギュレーションされました。
- RNAseqは、核あたり少数の差次的発現遺伝子(DEG)を特定しましたが、多重比較補正後の有意な所見はありませんでした。
- OCD診断は、遺伝子発現または共発現遺伝子モジュールを効果的にモデル化しませんでした。
結論:
- このパイロット研究では、強迫性障害における研究された視床核における有意な差次的RNA発現の証拠はほとんど見られませんでした。
- これらの発見は、視床の遺伝子発現の変化が、他の脳領域での発見とは対照的に、OCD病態生理の主な原因ではない可能性を示唆しています。
