  • CRISPR-Cas9キルスイッチは、生存可能な遺伝子改変微生物(CFU)を大幅に減少させたが、ターゲット遺伝子の存在量が高く、完全なDNAが存在することを示唆していた。
  • 遺伝子存在量による高い逃避率(10-1.6~10-1.0)が、低いCFU率(10-6.2)とは対照的に観察された。
  • 遺伝子改変微生物のDNAは、少なくとも1時間は細胞内で大部分が完全でDNase耐性を示したが、河川水中で数日かけて分解された。

結論:

  • CRISPR-Cas9キルスイッチはGEMの増殖を効果的に抑制するが、検出可能なDNAを排除するわけではない。
  • 完全なGEM DNAは生物学的封じ込め後も持続し、リスクをもたらし、モニタリングを複雑にする。
  • 環境リスクを軽減するために、生物学的封じ込めがGEMとそのDNAに与える影響を完全に理解するためには、さらなる研究が必要である。
