Summary
We identified mouse Lampetins (lysosome-associated membrane proteins) and mapped their developmental expression. These proteins may play roles in cell interactions and lysosome function during tissue development.
Area of Science:
- Developmental Biology
- Cell Biology
- Molecular Biology
Background:
- Lysosome-associated membrane proteins (Lamp)-2 are found in lysosomes and on the plasma membrane.
- Lamp-2 proteins are implicated in cell adhesion and secretion of lysosomal hydrolases.
- The precise functions of Lamp-2 proteins, especially during development, require further elucidation.
Purpose of the Study:
- To isolate and characterize murine Lampetins (Lamp)-2 homologues.
- To investigate the developmental expression patterns of murine Lamp-2 transcripts.
- To correlate expression patterns with potential protein functions during morphogenesis.
Main Methods:
- Isolation and characterization of murine Lamp-2 transcripts.
- Analysis of transcript expression patterns during mouse development using molecular techniques.
- Detection of Lamp-2 protein in specific tissues.
Main Results:
- Murine Lamp-2 transcripts (m-Lamp-2a, -2b, -2c) were identified and characterized.
- Distinct expression patterns of m-Lamp-2 transcripts were observed in neural crest, mesoderm, and endoderm-derived tissues.
- m-Lamp-2a and -2c showed specific expression in tissues undergoing epithelial-mesenchymal interactions and branching morphogenesis.
Conclusions:
- All m-Lamp-2 transcripts are present in tissues undergoing apoptosis, suggesting a role in phagolysosome function.
- Lamp-2 expression patterns indicate involvement in cell-cell and cell-extracellular matrix interactions during development.
- Lamp-2 proteins likely have tissue-specific roles in lysosome function during morphogenesis.
