Summary

Estrogen depletion impairs spatial working memory in mice, but not spatial reference memory. NMDA receptor antagonism equally affected performance in both intact and estrogen-depleted mice, suggesting estrogen

Area of Science:

  • Neuroscience
  • Behavioral Neuroscience
  • Endocrinology

Background:

  • Estrogen and NMDA receptor antagonists influence the hippocampus CA1 region and memory functions.
  • Estrogen's role in spatial memory, particularly in relation to NMDA receptor activity, requires further investigation.

Purpose of the Study:

  • To investigate the interactive effects of estrogen depletion and NMDA receptor antagonism on spatial memory in mice.
  • To determine if NMDA receptor antagonists mediate the effects of estrogen on spatial memory.

Main Methods:

  • Ovariectomy or sham operations were performed on mice, followed by a recovery period.
  • Mice received intraperitoneal injections of vehicle or the NMDA receptor antagonist CPP (2, 5, 10 mg/kg).
  • Spatial reference memory (water maze), spatial working memory (radial arm maze), and locomotive activity (Y-maze) were assessed.

Related Experiment Videos

Main Results:

  • No significant differences in spatial reference memory or locomotive activity were observed between ovariectomized and sham-operated mice.
  • Ovariectomized mice exhibited impaired spatial working memory compared to intact mice in the radial arm maze.
  • CPP treatment dose-dependently impaired performance in both the water maze and radial arm maze for all groups, with no effect on Y-maze activity.

Conclusions:

  • Short-term estrogen deprivation impairs spatial working memory but not spatial reference memory.
  • The observed impairment in spatial working memory due to estrogen depletion is likely not mediated by NMDA receptors.
  • NMDA receptor antagonism affects spatial memory tasks independently of estrogen status.
