Abstract:
A method for graphically analyzing the pH dependence of enzyme activity is presented. Linear plots can be made to test the applicability of the assumed models. For three simple cases, if the pH dependence curves are measured at two substrate concentrations, it is possible to evaluate ion dissociation constants from the linear plots. The method would be useful for studies of pH effects on enzyme activities if applied complementarily to other methods.
