Background:

  • The T-cell receptor (TCR) V beta 8 (Vβ8) gene segment is frequently utilized by antigen-specific T-cells restricted by MHC class II molecules I-Ad and I-Ed.
  • Previous studies confirmed the in vivo importance of Vβ8 in responses to I-Ad/Ed-restricted antigens using a Vβ8 depletion assay.

Purpose of the Study:

  • To investigate whether Vβ8 is also dominantly used by alloreactive T-cells that recognize the same MHC class II molecules (I-Ad/Ed).
  • To compare TCR Vβ8 usage between antigen-specific and alloreactive T-cell populations.
