Summary

Lactobacilli strains exhibit varied citrate utilization, with some using it as an energy source and others co-metabolizing it with glucose. All studied strains possess a citrate transport system inducible by citrate.

Area of Science:

  • Microbiology
  • Food Science
  • Biochemistry

Background:

  • Lactobacilli are crucial in fermented foods and human health.
  • Understanding their metabolic capabilities, like citrate utilization, is key to optimizing fermentation processes and probiotic applications.
  • Citrate metabolism in lactobacilli can influence flavor profiles and nutrient availability.

Purpose of the Study:

  • To investigate and differentiate citrate utilization patterns among various lactobacilli strains.
  • To identify strains capable of using citrate as a sole energy source versus those that co-metabolize it.
  • To explore the inducibility and characteristics of the citrate transport system in lactobacilli.

Main Methods:

  • Culturing lactobacilli in Kempler and McKay and calcium citrate media, with and without glucose.

Related Experiment Videos

  • Analyzing growth and metabolic products to determine citrate utilization pathways.
  • Conducting citrate transport experiments and dot blot assays to investigate the citrate permease system.
  • Main Results:

    • Calcium citrate medium with glucose effectively distinguished citrate-fermenting lactobacilli.
    • Lactobacillus rhamnosus, Lactobacillus zeae, and Lactobacillus plantarum utilized citrate as their sole energy source.
    • Other strains co-metabolized glucose and citrate, with variable induction levels of the citrate transport system.
    • No equivalent plasmid coding for citrate permease was detected in the studied lactobacilli.

    Conclusions:

    • Citrate utilization is a strain-specific trait in lactobacilli, impacting their metabolic versatility.
    • The presence of an inducible citrate transport system is common, but its efficiency varies.
    • Citrate metabolism can contribute to the production of aroma compounds in fermented products.
