    Summary

    Thymosin, a calf thymus extract, boosts T-cell rosettes in patients with immune deficiencies. This immune reconstitution may predict patient response to thymosin therapy.

    Background:

    • Primary immunodeficiency diseases and viral illnesses can impair cellular immunity.
    • T-cell rosettes are a marker of T-lymphocyte function.

    Purpose of the Study:

    • To investigate the effect of thymosin on T-cell rosette formation.
    • To assess the clinical utility of thymosin in patients with cellular immunodeficiency.

    Main Methods:

    • In vitro incubation of lymphocytes with thymosin and sheep erythrocytes.
    • In vivo administration of thymosin to a patient with thymic hypoplasia.
    • Monitoring of T-cell rosette percentages, clinical status, and skin test reactivity.

    Main Results:

    • Thymosin increased T-cell rosettes in vitro in patients with primary immunodeficiency diseases or viral illness.
    • A patient with thymic hypoplasia showed clinical improvement and normalization of T-cell rosettes during in vivo thymosin therapy.
    • Delayed hypersensitivity skin tests became positive during thymosin treatment.

    Conclusions:

    • Thymosin may partially restore cellular immunity in patients with immunodeficiencies.
    • Increased T-cell rosettes after in vitro thymosin incubation may predict in vivo therapeutic response.
