    Summary

    Patients with low or absent immunoglobulin A (IgA) showed normal immune cell function. Their B cells could still produce IgA, though some showed suppressed IgA production when stimulated with PWM.

    Background:

    • Selective IgA deficiency is a primary immunodeficiency.
    • Understanding B cell function in IgA deficiency is crucial for immune response research.

    Purpose of the Study:

    • To investigate the functional capacity of B cells and lymphocyte responses in patients with IgA deficiency.
    • To assess the in vitro differentiation and IgA secretion of B cells from these patients.

    Main Methods:

    • Compared immune cell parameters (E-RFC, lymphocyte DNA synthesis) in IgA-deficient patients and normal subjects.
    • Assessed B cell populations and IgA-bearing lymphocytes.
    • Evaluated lymphocyte DNA synthesis induced by various mitogens and antibodies.
    • Quantified IgA and IgG secretion from cultured B cells stimulated with pokeweed mitogen (PWM).

    Main Results:

    • Patients exhibited normal E-RFC counts and lymphocyte DNA synthesis.

    • PWM's differential effect on IgA B cell differentiation suggests complex regulatory mechanisms in IgA deficiency.
