Summary

Thrice-weekly buprenorphine dosing is as effective as daily dosing for opioid agonist maintenance treatment. This study found comparable retention and reduced illicit drug use with both buprenorphine regimens.

Area of Science:

  • Pharmacology
  • Addiction Medicine
  • Clinical Trials

Background:

  • Buprenorphine is a viable alternative to methadone for opioid agonist maintenance treatment.
  • Optimizing buprenorphine dosing schedules, such as thrice-weekly administration, could enhance patient adherence and treatment accessibility.

Purpose of the Study:

  • To compare the efficacy of daily versus thrice-weekly sublingual buprenorphine in opioid-dependent patients.
  • To assess treatment retention, illicit drug use, and medication compliance between the two dosing groups.

Main Methods:

  • A 12-week randomized, double-blind study involving 92 opioid-dependent patients.
  • Patients received either daily or thrice-weekly sublingual buprenorphine after a 3-day induction period.
  • Outcomes measured included retention rates, urine toxicology, and self-reported illicit drug use.

Related Experiment Videos

Main Results:

  • No significant baseline differences were observed between the daily and thrice-weekly buprenorphine groups.
  • Treatment retention was similar, with 71% in the daily group and 77% in the thrice-weekly group.
  • Both groups showed significant reductions in opioid-positive urine tests, with no significant differences in self-reported heroin use or medication compliance.

Conclusions:

  • Thrice-weekly sublingual buprenorphine is comparable in efficacy to daily dosing for opioid agonist maintenance treatment.
  • The findings support the use of thrice-weekly buprenorphine dosing in diverse clinical settings.
  • This dosing schedule may improve treatment accessibility and patient convenience.
