    Area of Science:

    • Psychiatry and Pharmacology
    • Clinical Neuroscience

    Background:

    • Maintenance therapy is crucial for managing chronic schizophrenia.
    • Evaluating novel antipsychotics against established treatments and placebo is essential.

    Purpose of the Study:

    • To assess the efficacy of thioridazine (THI) and pimozide (PIM) as maintenance treatments for chronic schizophrenic female outpatients.
    • To establish the sensitivity of the experimental design using placebo (PL) as a comparator.

    Main Methods:

    • A 6-month, double-blind, placebo-controlled clinical trial.
    • Participants received daily doses of THI (≤375 mg/day), PIM (≤16 mg/day), or PL.
    • Treatment efficacy was evaluated using the Brief Psychiatric Rating Scale (BPRS) and social adjustment ratings.

    Main Results:

    • THI and PIM showed significant maintenance treatment effects compared to PL.
    • Treatment failure rates were 56% for PL, 14% for THI, and 17% for PIM.
    • Improvements in global impression were noted in both THI and PIM groups.

    Conclusions:

    • Thioridazine and pimozide are effective maintenance therapies for chronic schizophrenia.
    • The study design was sensitive enough to detect treatment differences.
    • While BPRS and social adjustment ratings showed some differences, they were generally insensitive to treatment effects.
