Nidation inhibition by simple ergoline derivatives.

K Rezábek, J M Cassady, H G Floss

    Journal of Pharmaceutical Sciences

    Summary

Four ergoline compounds were tested for their ability to inhibit nidation in rats. Their effectiveness in blocking nidation paralleled their prolactin-inhibiting activity, suggesting a link between these functions.

    Area of Science:

    • Pharmacology
    • Reproductive Biology
    • Endocrinology

    Background:

    • Ergoline derivatives are known for diverse biological activities.
    • Prolactin plays a crucial role in reproductive processes, including implantation.
    • Inhibition of nidation is a potential mechanism for fertility control.

    Purpose of the Study:

    • To investigate the nidation inhibitory effects of specific ergoline-type compounds.
    • To correlate the nidation inhibitory activity with prolactin-inhibiting properties.

    Main Methods:

    • Administration of four ergoline derivatives to rats.
    • Assessment of nidation inhibition.
    • Evaluation of prolactin-inhibiting activity.

    Main Results:

    • Elymoclavine and its derivatives demonstrated nidation inhibitory effects in rats.
    • The degree of nidation inhibition correlated directly with prolactin-inhibiting activity.
    • N-methyl-6,7-secoelymoclavine also exhibited similar effects.

    Conclusions:

    • Ergoline-type compounds possess the ability to inhibit nidation in rats.
    • Prolactin inhibition is a likely mechanism underlying the observed anti-nidation effects.
    • These findings suggest potential applications for ergoline derivatives in reproductive health.
