Improving Translational Accuracy02:07

Improving Translational Accuracy

15.1K
Base complementarity between the three base pairs of mRNA codon and the tRNA anticodon is not a failsafe mechanism. Inaccuracies can range from a single mismatch to no correct base pairing at all. The free energy difference between the correct and nearly correct base pairs can be as small as 3 kcal/ mol. With complementarity being the only proofreading step, the estimated error frequency would be one wrong amino acid in every 100 amino acids incorporated. However, error frequencies observed in...

Nonsense-mediated mRNA Decay02:27

Nonsense-mediated mRNA Decay

11.9K
The Upf proteins that carry out nonsense-mediated decay (NMD) are found in all eukaryotic organisms, including humans. Each protein has an individual role, but they need to work in collaboration. Upf1 is an ATP-dependent RNA helicase that unwinds the RNA helix. Because Upf1 can unwind any RNA, Upf2 and Upf3 are required to help Upf1 discriminate between nonsense and normal mRNAs.
Usually, Upf3 binds to an Exon Junction Complex (EJC) at mRNA splice sites. If a ribosome fully translates the mRNA,...

piRNA - Piwi-interacting RNAs02:57

piRNA - Piwi-interacting RNAs

7.7K
PIWI-interacting RNAs, or piRNAs, are the most abundant short non-coding RNAs. More than 20,000 genes have been found in humans that code for piRNAs while only 2000 genes have been found for miRNAs. piRNAs can act at the transcriptional and post-transcriptional levels and have a vital role in silencing transposable elements present in germ cells. They are also involved in epigenetic silencing and activation. Previously, they were thought to function only in germ cells but new evidence suggests...

Summary
This summary is machine-generated.

Premature adrenarche, early pubic hair growth, typically has no long-term effects. However, some girls may develop functional ovarian hyperandrogenism and hyperinsulinemia later in life.

Area of Science:

  • Pediatric Endocrinology
  • Reproductive Endocrinology

Context:

  • Premature adrenarche is defined as the appearance of pubic hair before age 8 in girls and 9 in boys.
  • It is often associated with accelerated growth and bone maturation but typically does not affect pubertal onset or final height.

Purpose:

  • To review the diagnosis and management of premature adrenarche.
  • To highlight emerging concerns regarding the long-term outcomes in girls with this condition.

Summary:

  • Diagnosis involves excluding other forms of hyperandrogenism.
  • While often benign, recent data suggest girls with premature adrenarche may have an increased risk of functional ovarian hyperandrogenism.
  • Hyperinsulinemia is frequently observed in these patients, potentially linked to androgen excess.

Impact:

  • Emphasizes the need for long-term follow-up in girls diagnosed with premature adrenarche.
  • Suggests a potential link between premature adrenarche, hyperinsulinemia, and functional ovarian hyperandrogenism, possibly stemming from common early origins.
