Summary

Microstructural tissue alterations occur in the putamen and thalamus in cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL). These changes, particularly in the thalamus, may indicate white matter damage and impact cognitive function.

Background:

  • Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy (CADASIL) is characterized by white matter changes and lacunar infarcts.
  • Previous studies indicated microstructural alterations in white matter, but subcortical gray matter changes outside lesions were unknown.

Purpose of the Study:

  • To investigate microstructural tissue alterations in the putamen and thalamus of CADASIL patients.
  • To determine if these changes occur outside of visible lacunar infarcts.

Main Methods:

  • Diffusion Tensor Imaging (DTI) was used to assess cerebral microstructure in 20 CADASIL patients and 12 controls.
  • Measurements included the trace of the diffusion tensor [Tr(D)] and an anisotropic diffusion index within the putamen and thalamus.

  • Patients with CADASIL showed increased Tr(D) and decreased anisotropy in the putamen and thalamus.
