[Effect of protein expression on antisense c-myc and PCNA for vascular smooth muscle cells]
1Department of Vascular Surgery, Renji Hospital, Shanghai Second Medical University, Shanghai 200001, China.
Objective:
To study the methods of inhibiting proliferation of vascular smooth muscle cells (VSMCs), we observed the change of protein expression after using antisense c-myc, PCNA for VSMCs.
Methods:
The sense, antisense, and mismatched oligomers for c-myc or PCNA were individually delivered to the cultured VSMCs. Using Western blot, immunohistochemistry assay and image analysis system, we observed the change of c-myc and PCNA protein expression in the VSMCs.
Results:
Antisense c-myc, PCNA at concentration of 10 micromol/L can successfully inhibit the expression of c-myc, PCNA protein in the VSMCs (P < 0.05), but the sense and the mismatched do not have this effect (P > 0.05).
Conclusion:
The data suggested that the antisense c-myc, PCNA could attenuate the protein expression of c-myc, PCNA, and it is an effective method for the inhibition of VSMCs hyperplasia.
Insights
Antisense c-myc and PCNA effectively inhibit vascular smooth muscle cell proliferation by reducing protein expression. This offers a promising method for controlling VSMC hyperplasia.
Area of Science:
- Molecular Biology
- Cell Biology
- Cardiovascular Research
Context:
- Vascular smooth muscle cell (VSMC) proliferation is a key factor in cardiovascular diseases like atherosclerosis and restenosis.
- Understanding the molecular mechanisms regulating VSMC growth is crucial for developing targeted therapies.
Purpose:
- To investigate the efficacy of antisense oligomers targeting c-myc and PCNA in inhibiting VSMC proliferation.
- To analyze the impact of antisense c-myc and PCNA on the protein expression levels within VSMCs.
Summary:
- Antisense oligomers for c-myc and PCNA, at a concentration of 10 micromol/L, significantly inhibited the expression of these proteins in cultured VSMCs (P < 0.05).
- Sense and mismatched oligomers did not demonstrate a significant inhibitory effect on c-myc and PCNA protein expression (P > 0.05).
- Western blot, immunohistochemistry, and image analysis confirmed the downregulation of c-myc and PCNA proteins by antisense treatment.
Impact:
- The findings suggest that antisense c-myc and PCNA are effective in attenuating protein expression, presenting a viable strategy for inhibiting VSMC hyperplasia.
- This research provides a potential therapeutic avenue for managing conditions characterized by excessive VSMC growth.
