We developed a simple method to link semiconductor quantum dots (QDs) to antibodies using avidin. This allows for sensitive detection of protein toxins and potential for multianalyte detection.
Area of Science:
- Bioinorganic chemistry
- Nanotechnology
- Immunotechnology
Background:
- Semiconductor nanocrystal quantum dots (QDs) offer unique luminescent properties for bioimaging and sensing.
- Antibody conjugation to nanomaterials is crucial for targeted detection but often involves complex procedures.
Purpose of the Study:
- To develop a facile method for conjugating luminescent quantum dots (QDs) to antibodies.
- To characterize the resulting bioinorganic conjugates.
- To evaluate their utility in fluoroimmunoassays for protein toxin detection.
Main Methods:
- Electrostatic self-assembly of avidin onto negatively charged CdSe-ZnS core-shell QDs modified with dihydrolipoic acid.
- Conjugation of biotinylated antibodies to the avidin-QD complex.
- Fluoroimmunoassays for detecting staphylococcal enterotoxin B and cholera toxin.
Main Results:
- Stable bioinorganic conjugates of QDs and antibodies were successfully prepared.
- The avidin bridge facilitated strong adsorption of avidin to the QD surface.
- The QD-antibody conjugates effectively detected target protein toxins in fluoroimmunoassays.
Conclusions:
- A straightforward and effective method for QD-antibody conjugation using avidin has been established.
- These conjugates serve as versatile immunoassay reagents.
- The approach holds promise for developing multiplexed or multianalyte detection systems.

Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding.
