[(A1-D-alanine) insulin (author's transl)].

R Geiger, K Geisen, H D Summ

    Hoppe-Seyler'S Zeitschrift Fur Physiologische Chemie
    |October 1, 1975
    PubMed
    Summary

Replacing A1-glycine with D-alanine in porcine insulin maintained blood sugar lowering activity. This D-alanine insulin analogue showed enhanced glucose uptake and receptor binding compared to the L-alanine version.

    Area of Science:

    • Biochemistry
    • Endocrinology
    • Pharmacology

    Context:

    • Investigating insulin analogues to enhance therapeutic properties.
    • Understanding structure-activity relationships in peptide hormones.

    Purpose:

    • To synthesize and characterize [A1-D-Alanine]insulin and [A1-L-Alanine]insulin.
    • To compare the biological activity and receptor binding of these analogues to native porcine insulin.

    Summary:

    • Substitution of A1-glycine with L-alanine in porcine insulin decreased biological activity.
    • [A1-D-Alanine]insulin demonstrated equivalent blood glucose-lowering effects and increased glucose uptake in rat diaphragm compared to native insulin.
    • Receptor binding studies showed decreased affinity for [A1-D-Alanine]insulin compared to native insulin, but higher affinity than the L-alanine analogue.

    Impact:

    • Identifies [A1-D-Alanine]insulin as a potential therapeutic agent with preserved or enhanced activity.
    • Provides insights into the role of the A1 position in insulin receptor interactions.
    • Contributes to the development of novel insulin derivatives for diabetes management.
