Electrophoretic mobilities of beta-blocker drugs were measured in mixed solvents using capillary electrophoresis. A mathematical model accurately predicted these mobilities, showing low mean percentage errors for practolol, timolol, and propranolol.
Area of Science:
- Analytical Chemistry
- Physical Chemistry
Background:
- Beta-blocker drugs like practolol, timolol, and propranolol are crucial in cardiovascular treatment.
- Understanding their behavior in different solvent systems is essential for pharmaceutical analysis.
Purpose of the Study:
- To measure the electrophoretic mobilities of three beta-blocker drugs in various water-organic solvent mixtures.
- To evaluate a mathematical model predicting drug mobility based on solvent composition.
Main Methods:
- Capillary electrophoresis was employed to determine electrophoretic mobilities.
- Binary (water-methanol, water-ethanol) and ternary (water-methanol-ethanol) solvent systems were used with acetic acid/sodium acetate buffer.
- A mixture response surface method was utilized to model mobility as a function of solvent composition.
Main Results:
- Highest electrophoretic mobilities were observed in pure aqueous buffers, lowest in ethanolic buffers.
- The mathematical model showed good accuracy, with mean percentage errors (MPE) between 0.9-2.6% for binary mixtures and ~2.7% for ternary mixtures.
- Calculated MPE values were within experimental relative standard deviation, indicating acceptable error.
Conclusions:
- The developed mathematical model accurately predicts the electrophoretic mobilities of beta-blockers in mixed aqueous-organic solvents.
- This predictive capability is valuable for optimizing capillary electrophoresis methods in pharmaceutical analysis.
