Summary

T cell adhesion primes T cells for antigen detection by increasing intracellular calcium and phosphatidylinositol 4,5-bisphosphate levels, enhancing T cell receptor (TCR) signaling. This adhesion-dependent priming is crucial for sensitive antigen recognition by T cells.

Area of Science:

  • Immunology
  • Cellular Biology
  • Biochemistry

Background:

  • T cell receptor (TCR) signaling is critical for adaptive immunity.
  • Physiological conditions require T cell adhesion for antigen detection and TCR signaling initiation.
  • The role of T cell adhesion in modulating TCR signaling remains incompletely understood.

Purpose of the Study:

  • To investigate the mechanisms by which T cell adhesion enhances TCR signaling.
  • To determine the impact of T cell adhesion on calcium (Ca2+) signaling and phosphatidylinositol 4,5-bisphosphate (PIP2) levels.
  • To address the controversy regarding the minimum valency of TCR ligands required for T cell activation.

Main Methods:

  • Analysis of Ca2+ signaling in adherent versus suspended T cells.

Related Experiment Videos

  • Quantification of intracellular Ca2+ stores and plasma membrane PIP2 levels.
  • Stimulation of T cells with monomeric MHC-peptide complexes on different substrates.
  • Main Results:

    • T cell adhesion increases intracellular Ca2+ stores and plasma membrane PIP2.
    • Adhesion enhances TCR-induced Ca2+ signaling in various T cell types.
    • Specific monomeric MHC-peptide complexes trigger Ca2+ responses only after T cell adhesion.
    • Adhesion-induced priming occurs with both artificial substrates and dendritic cells.

    Conclusions:

    • T cell adhesion acts as a priming mechanism that enhances TCR signaling sensitivity.
    • Adhesion-mediated priming resolves the valency issue for TCR ligand activation.
    • This phenomenon likely contributes to the high sensitivity of T cell antigen detection in lymphoid organs.
