Summary

Fibroblast growth factor 8 (Fgf8) and Fgf24 are essential for posterior mesoderm development in zebrafish. Co-inhibition of fgf8 and fgf24 severely disrupts mesoderm formation, highlighting their synergistic role.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • Fibroblast growth factor (Fgf) signaling is crucial for vertebrate embryonic development, particularly posterior mesoderm formation.
  • While Fgf8 is essential for mesoderm development in mice, zebrafish fgf8 mutants exhibit only mild posterior mesodermal defects, suggesting other Fgf ligands are involved.

Purpose of the Study:

  • To identify and characterize novel Fgf ligands involved in zebrafish posterior mesoderm development.
  • To elucidate the functional interaction between Fgf8 and other Fgf family members in this process.

Main Methods:

  • Identification and expression analysis of fgf8-related genes in zebrafish.
  • Morpholino-based gene inactivation to study gene function.
  • Analysis of gene interactions with known posterior mesoderm T-box transcription factors (no tail, spadetail).

Related Experiment Videos

Main Results:

  • Fgf24 is co-expressed with Fgf8 in mesodermal precursors during gastrulation.
  • Inhibition of fgf24 alone has no effect, but combined inhibition with fgf8 blocks posterior mesoderm formation.
  • Fgf8 and Fgf24 interact with no tail and spadetail transcription factors.
  • fgf24 is required for pectoral fin development; its inhibition leads to finless fish.

Conclusions:

  • Fgf8 and Fgf24 function redundantly and are jointly required for posterior mesoderm development in zebrafish.
  • These Fgf signaling pathways interact with T-box transcription factors to regulate posterior mesoderm formation.
  • Fgf24 plays a specific role in pectoral fin development.
