Background:

  • Endothelin-1 (ET-1) is a potent vasoconstrictor with roles in blood pressure regulation.
  • The effect of acute volume expansion on ET-1 release is not fully understood.

Purpose of the Study:

  • To investigate the hypothesis that acute volume expansion triggers endothelin-1 release.
  • To determine if saline infusion alters plasma endothelin-1 levels in rats.

Main Methods:

  • Conscious Wistar rats and spontaneously hypertensive rats underwent bolus intravenous saline infusion.
  • Mean arterial pressure and heart rate were monitored.
  • Plasma endothelin-1 levels were measured before and after saline infusion.

Main Results:

  • Saline infusion did not significantly alter mean arterial pressure or heart rate.
  • Plasma endothelin-1 levels increased significantly in both Wistar rats (42%) and spontaneously hypertensive rats (61%) one minute post-infusion.
  • In Wistar rats, endothelin-1 levels rose from 0.68 +/- 0.13 to 1.19 +/- 0.17 fmol/mL (P <.0001).

Conclusions:

  • Acute volume expansion via rapid saline infusion effectively increases plasma endothelin-1 levels.
  • Endothelin-1-induced vasoconstriction may serve as a compensatory mechanism against the increased circumferential stretch resulting from volume expansion.
