Insights
Determining optimal drug dosage in children is complex due to growth and developmental changes. This review discusses factors influencing pediatric drug dosing and administration for herpesvirus infections.
Area of Science:
- Pharmacology
- Pediatric Medicine
- Infectious Diseases
Background:
- Optimizing drug dosage is crucial for effective treatment, especially in pediatric patients.
- Pediatric drug dosing is challenging due to pharmacokinetic variability, growth, and organ development.
- Lack of specific pediatric pharmacokinetic studies complicates accurate dosing.
Purpose of the Study:
- To review factors and misconceptions in determining optimal drug dose and administration routes for pediatric populations.
- To discuss pharmacokinetics, appropriate doses, and preferred routes for specific antiviral drugs used in children.
Main Methods:
- Literature review of factors affecting pediatric drug dosing.
- Analysis of pharmacokinetic data for selected antiviral medications.
- Discussion of administration routes and dosages for treating herpesvirus infections in children.
Main Results:
- Identified key factors influencing pediatric drug pharmacokinetics and dosing.
- Summarized appropriate doses and preferred routes for aciclovir, ganciclovir, famciclovir, foscarnet, and cidofovir.
- Highlighted common misconceptions in pediatric drug administration.
Conclusions:
- Accurate pediatric drug dosing requires careful consideration of individual patient factors and drug-specific pharmacokinetics.
- Optimized dosing strategies are essential for effective herpesvirus treatment in children.
- Further research on pediatric pharmacokinetics is needed to improve treatment outcomes.
Abstract:
Effective treatment requires administration of optimal drug dose. In paediatric patients this may be difficult to achieve due to lack of pharmacokinetic studies, administration of small doses, changes in body composition, continuous growth, and development and maturation of organs and systems. This article reviews the factors and misconceptions associated with determining optimal drug dose and route of administration in different paediatric populations. The pharmacokinetics, appropriate doses and preferred routes of administration for aciclovir, ganciclovir, famciclovir, foscarnet and cidofovir, for treating herpesvirus infections in the paediatric population, are discussed.
