Summary

Prostate cancer in dogs and humans can cause new bone formation at metastatic sites. Canine prostate tissue stimulates osteoblast activity through an endothelin-dependent pathway, not involving PTHrP or prostaglandins.

Background:

  • Bone metastases are common in advanced prostate cancer in humans and dogs, characterized by new bone formation (osteoblastic metastases).
  • Unlike many cancers that cause bone destruction, prostate cancer promotes bone formation, a mechanism not fully understood.
  • Prostate cancer's induction of bone formation is hypothesized to involve paracrine actions of growth factors from prostate epithelial cells.

Purpose of the Study:

  • To investigate the mechanisms underlying prostate-induced new bone formation.
  • To develop an in vitro system to study prostate-stimulated osteoblast function, complementing existing in vivo models.

  • Utilized an in vitro system with cultured rat calvaria treated with proteins from normal canine prostate tissue.

    • This study establishes a reliable in vitro assay for studying prostate-mediated osteoblast activation.
