Summary

The 3G5 ganglioside antigen is newly found on melanocytes and skin tumors. This antigen shows high expression in benign and malignant melanocytic tumors but is minimally expressed in normal melanocytes.

Background:

  • The 3G5 ganglioside antigen (3G5 antigen) is known to be expressed on various cell types.
  • Its expression on melanocytes and common skin tumors has not been previously reported.

Purpose of the Study:

  • To investigate the expression of the 3G5 antigen in normal skin and various skin tumors, including melanocytic nevi, malignant melanoma, basal cell carcinoma, and seborrheic keratosis.
  • To determine if 3G5 antigen expression differs between normal melanocytes and tumor-associated melanocytes.

Main Methods:

  • Analysis of 148 skin cryosections from normal skin and 45 skin tumors using triple fluorescence technique.
  • Utilized monoclonal antibody 3G5, DNA fluorochrome, and anti-melanocytic antibody A103 (Anti-Melan-A).

  • 3G5 antigen expression was detected in epidermal melanocytes of normal skin (14.8% positive).
  • High expression of 3G5 antigen was observed in melanocytic nevi (72.2%), primary melanomas (83.9%), and melanoma metastases (82.5%).
  • Basal cell carcinomas and seborrheic keratoses showed no 3G5 antigen expression on tumor cells.

Conclusions:

  • This study reports for the first time the expression of the 3G5 antigen in melanocytes.
  • The findings indicate significantly higher 3G5 antigen expression in aggregated melanocytes of benign and malignant tumors compared to singular melanocytes in normal epidermis.
  • This differential expression suggests a distinct biological state associated with 3G5 antigen in melanocytic neoplasms.
