Intervertebral disc degeneration risk may be influenced by IL-1 gene variations. Occupational physical load effects on disc degeneration appear modified by these IL-1 gene polymorphisms.

Purpose of the Study:

  • To investigate the association between IL-1 gene locus polymorphisms and lumbar disc degeneration.
  • To determine if occupational physical load modifies the impact of these polymorphisms on disc degeneration.

Main Methods:

  • Genotyping was performed on 133 middle-aged men from diverse occupations (machine drivers, carpenters, office workers).
  • Lumbar spine MRI evaluated signs of degeneration, including decreased nucleus pulposus signal intensity, disc bulges, and reduced disc height.
  • Degeneration was assessed in discs from L2/L3 to L5/S1 levels.

  • Carriers of IL-1alphaT or IL-1betaT alleles showed increased odds ratios for disc bulges (2.4 and 1.9, respectively).
  • The IL-1alpha TT genotype was associated with a >3-fold increased risk of disc bulges compared to the CC genotype.

  • Polymorphisms in the IL-1 gene cluster may influence an individual's susceptibility to disc degeneration.
