Abstract:
IRSs (insulin receptor substrates) are substrates for both insulin and IGF-I receptors. Insulin and IGF-I are known to be bone anabolic factors. IRS-1 deficient mice exhibited severe low turnover osteopenia with decreased bone anabolic and catabolic action, while IRS-2 deficient mice showed osteopenia with decreased anabolic and increased catabolic action. IRS-1 signaling plays an important role for maintaining bone turnover while IRS-2 signaling maintains anabolic balance in osteoblasts.
