Summary

Following bone fracture and soft tissue injury, local lymph nodes enlarge and activate immune cells from the blood. The exact signaling mechanism triggering this response in lymph nodes remains unknown.

Background:

  • Clinical studies show enlarged lymph nodes (LN) and dilated lymphatics in patients with post-traumatic edema.
  • Previous observations suggest a link between tissue injury and lymphatic system response.

Purpose of the Study:

  • To investigate changes in rat lymph nodes draining a tibial fracture and soft tissue injury.
  • To analyze lymph node weight, cell counts, phenotypes, cytokine expression, and mitogen responsiveness.

Main Methods:

  • Rats with tibial fractures and soft tissue injuries were studied.
  • Parameters assessed included lymph node weight, cell number, flow cytometry, immunohistochemistry, and cytokine analysis.

  • Decreased frequencies of ED1 macrophages and OX8 cytotoxic cells were observed.

Conclusions:

  • Local lymph nodes react to internal wounds by mobilizing and activating blood-borne cells.
  • The molecular signals initiating this lymph node reaction are currently unknown.
  • Lymph nodes appear prepared for antigen influx, reacting with increased cell number and activation, but the lack of auto-antigen immunization requires further study.
